Improved vitality and health: A landmark human study on NMN supplementation

Research consensus indicates that high NAD levels in humans are beneficial, promote longevity, and help prevent disease. Therefore, anti-aging researchers are continuously exploring strategies to increase NAD levels as we grow older.
Supplementation with Nicotinamide Mononucleotide (NMN), a natural molecule that our bodies convert into NAD, may be one such method. Several studies correlate NMN intake with increased NAD levels, although correlation does not guarantee causation. Only animal studies had confirmed the effect of NMN supplementation on increasing NAD concentrations until this groundbreaking human study.
An international team of researchers tested the efficacy and safety of NMN supplementation in a clinical human study, published in GeroScience Journal in December 2022. The comprehensive, multi-goal study examined how different NMN doses affected NAD concentrations, NMN tolerability and safety, and its clinical efficacy using various assessments.
The gold-standard randomized, double-blind, placebo-controlled, dose-dependent study involved 80 middle-aged (40-65 years) healthy adults. They were divided into three experimental groups and one placebo control group, each receiving 300, 600, or 900 mg NMN orally daily.
This 60-day study involved thorough assessments, including safety and tolerability checks, NAD concentration measurement, endurance testing, "biological age" calculation, insulin resistance evaluation, and quality-of-life assessments.

The results indicated that NMN supplementation is safe and relatively well tolerated, with a significant increase in NAD concentration. NMN also improved physical endurance, reduced "biological age," and improved the participants' perceived quality of life and general health.
Interestingly, the study showed that a daily dose of 600 mg provided significant improvements over 300 mg, but increasing the dose to 900 mg provided no additional benefits. This study suggests that daily NMN supplementation may have a positive impact on overall health, well-being, aging, and endurance.
General information about NMN
NMN (nicotinamide mononucleotide), a naturally produced molecule and NAD precursor, offers several health benefits, especially for aging and longevity. Consuming NMN supplements, available as powder or capsules, may potentially increase NAD levels, leading to health improvements.
The main health benefits of NMN supplementation include:
- Improved energy metabolism and physical endurance, counteracting age-related decline in energy.
- Enhanced brain health and memory, promoting protection of brain cells and better cognitive function while mitigating neurodegenerative diseases.
- Improved heart health, contributing to blood pressure regulation, improved cholesterol levels, and prevention of blood clots to reduce the risk of cardiovascular disease.
- Positive effects on diabetes and obesity, through better insulin sensitivity, blood sugar control, and reduction in inflammation and oxidative stress.
- Skin rejuvenation, reduces signs of aging such as wrinkles and sagging skin through collagen production and improved hydration.
- Strengthened immune system, increases resistance to bacterial, viral, or fungal infections.
- Improved eye function, protects the eyes from damage caused by aging and light exposure, and improves visual quality.
